Colonel J H Rion resigned on 8 June, 1861, as he was not re-elected when the regiment mustered for Confederate service. On 8 July, following the participation in the Fort Sumter bombardment, Major C S Winder was assigned to the regiment.

The 6th South Carolina Infantry was one of the ten authorised for twelve months' service by the act of 17 December, 1860. The regiment was mustered in state service on 10 and 11 April, 1861, and in June, when it was reorganised and mustered in Confederate service, three companies, the Cedar Creek Rifle Company, the Fairfield Fencibles and the Little River Guard, elected not to enter Confederate service. Company A mustered in Confederate service on 12 June, companies B to G mustered in on 19 June with three new companies, H, I and K, joining about 21 June. The regiment was reorganised during April 1862.

The 6th South Carolina Infantry left Richmond, Virginia, on 20 July, and arrived at the Manassas battlefield on 21 July, too late in the day to be engaged.

On 25 July, 1861, the Army of the Potomac was reorganised and the 6th South Carolina Infantry was assigned to the Third Brigade, First Corps, Army of the Potomac, under the command of Brigadier General D R Jones.
